Tephrosia purpurea (L.) Pers., commonly known as "sarpunkha" and "wild indigo", is being used in traditional systems of medicine to treat liver disorders, spleen and kidney. In the present study, a validated High Performance Thin Layer Chromatography (HPTLC) method was established for the estimation of lupeol, ß-sitosterol and rotenone in various extracts of T. purpurea with the aim to see the effect of seasons on the quantity of aforesaid phytoconstituents. The plant material was collected in summer (April), rainy (August) and winter (December) during 2013-2014 from Lucknow, India. The method was validated in terms of precision, repeatability, specificity, sensitivity linearity and robustness. The method permits reliable quantification and showed good resolution on silica gel with toluene-ethyl acetate-formic acid (9:1:1 v/v/v) as mobile phase, and characteristic bands of ß-sitosterol, rotenone and lupeol were observed at Rf 0.38, 0.45 and 0.52, respectively. The content of aforesaid phytoconstituents varies from season to season and extract to extract. Our finding indicated that winter season (December) may not be appropriate for collection of T. purpurea for the preparation of therapeutic formulations because of the high content of rotenone, a known insecticide that is responsible for Parkinson's disease and associated with heart failure, fatty liver and liver necrosis.

BACKGROUND: Lymphatic filariasis (LF) is a parasitic disease that causes permanent disability (elephantiasis). Currently used antifilarial drugs are failing to control LF and there is resurgence in some areas. Looking for new antifilarial leads, we found that Calotropis procera plant parts have been used in traditional medicine for alleviating elephantiasis but the antifilarial activity is not known. OBJECTIVE: In the present study, the antifilarial activity of ethanolic extract (A001) and its hexane fraction (F001) of C. procera flowers was investigated using the human filarial parasite Brugia malayi. METHODS: A001 and F001 were tested for antifilarial activity using motility and 3-(4,5-dimethylthiazol-2- yl)-2,5 diphenyltetrazolium bromide (MTT) assays (in vitro) and in the rodent models B. malayi- Meriones unguiculatus and B. malayi-Mastomys coucha. In the rodent models, A001 and F001 were administered orally for 5 consecutive days, and the adult worm burden and course of microfilaraemia were determined. RESULTS: Both A001 and F001 showed microfilaricidal and macrofilaricidal activity in vitro. In animal models, A001 killed ~49-54% adult worms. In M. coucha model, F001 killed 12-60% adult worms in a dose (125-500 mg/kg) dependent manner; A001 and F001 suppressed microfilaraemia till days 91 and 35 post initiation of treatment, respectively. HPTLC revealed 0.61% lupeol, 0.50% ß-sitosterol and 1.50% triacontanol in F001. CONCLUSION: Flowers of C. procera have definite microfilaricidal and macrofilaricidal activities. Whether this activity is due to lupeol, ß-sitosterol and triacontanol found in the hexane fraction remains to be investigated. This is the first report on the antifilarial efficacy of flowers of the plant C. procera.

BACKGROUND: The quality of herbal medicine is determined by its secondary metabolites, which may vary according to growth, season and altitude etc. OBJECTIVE: We studied the variation in phytochemistry and biological activities of Thalictrum foliolosum (TF) roots collected from four sites at different altitudes. MATERIAL AND METHODS: The berberine content in different extracts of T. foliolosum roots collected from various altitudes was estimated using HPTLC. Total phenolic and flavonoid contents were determined using Folin-Ciocalteau reagent and aluminum chloride method respectively. The sensitivity of microbes for the extracts was studied using disc diffusion and the MIC was estimated using broth dilution method. Antioxidant capacity of the plant was studied using ß-carotene bleaching assay, lipid peroxidation assay using goat liver, reducing power assay and DPPH free radical scavenging activity. RESULTS: Berberine content varied inversely with altitude; while phenol and flavonoid content of TF increased at higher altitudes. All the TF extracts showed moderate to high activity against Candida albicans, Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa. Extracts with high berberine content were most effective against C. albicans and S. aureus and also showed relatively significant anti-lipid peroxidation, ß-carotene bleaching and reducing power. TF extracts with higher phenol and flavonoid content showed better scavenging of DPPH free radicals. Berberine was used as a standard in all the antioxidant and antimicrobial experiments performed. CONCLUSION: Thalictrum from lower elevations can be explored as an alternate source of berberine and the plant has high antioxidant and antimicrobial properties owing to its berberine content.

OBJECTIVE: Tephrosia purpurea (Linn·) Pers. is widely used in traditional medicine to treat liver disorders, febrile attacks, enlargement and obstruction of liver, spleen, and kidney. In the present study, investigations were carried out to determine the seasonal impact on the content of flavonoid glycosides and on antioxidant activities so as to identify the optimal time of harvesting. METHODS: The plant materials were collected in different seasons during 2013-2014. Air-dried, powdered plant materials were extracted with 95% ethanol and ethanol: water (1:1) by ultrasound-assisted extraction process. Their chemical composition in terms of total polyphenol and flavonoid contents (TPCs and TFCs) was determined using modified colorimetric Folin-Ciocalteu method and aluminum chloride colorimetric assay respectively. To determine the in vitro antioxidant activity, diphenyl-picryl hydrazyl (DPPH) radical-scavenging assay and total antioxidant capacity by phosphomolybdate antioxidant assay were carried out. High-performance liquid chromatography (HPLC)/photo-diode array (PDA) analysis was used to quantify the flavonoid glycosides in the samples collected in different seasons. Correlation studies were also carried out between antioxidant activities and TPCs. RESULTS: The highest TPC and TFC were found to be in the 95% ethanolic extract of the August sample and the lowest in the 50% hydro-alcoholic extract of the plant sample collected in winter season. It was observed that in both the assays used to determine the antioxidant activity, the 95% ethanolic extracts in all the seasons showed a higher activity than their respective 50% hydro-alcoholic extracts with an increase in activity as we go from cold to hot to rainy seasons. Based on correlation analysis, DPPH radical-scavenging activities as well as the spectrophotometrically measured phosphomolybdenum complex were also strongly correlated with TPC of the extracts. The most abundant flavonoid glycoside was quercetin-3-O-rhamnoglucoside in all the seasons. The content of all flavonoids was observed highest in the 95% ethanolic extract of the plant collected in August (TP-3). The 50% hydro-alcoholic extract of the plant collected in December (TP-6) showed the lowest amount of flavonoids and antioxidant activity. CONCLUSION: The findings of this study confirmed that the metabolism and production of flavonoids in T. purpurea are vigorously affected by seasonal factors. Significant differences were observed in the TPC, TFC and flavonoid glycoside composition in the 95% ethanolic as well as 50% hydro-alcoholic extracts of T· purpurea collected in different seasons. Since the 95% ethanolic extract of the August sample (TP-3), which also happens to be its flowering season, exhibited the highest TPC, TFC and antioxidant activities in both DPPH and phosphomolybdate assays as well as contained the highest content of all flavonoids, it could be recommended as the optimal season of harvesting T· purpurea with respect to its pharmaceutically important constituents, i.e., flavonoids.

A simple, sensitive and precise high-performance thin-layer chromatography method with densitometric detection was used for simultaneous determination of evernic (EV) and (+)-usnic acids (USN) in Usnea aciculifera (UA), U. ghattensis (UG), U. longissima (UL) and U. stigmatoides (US). This method was also validated according to the ICH guidelines. Separation and quantification was performed with the mobile phase toluene-1, 4-dioxane-formic acid (18:4.5:0.2, v/v/v) on silica gel 60F254 plates. The linearity for EV and USN was found in the 200-600 ng/band range. The limit of detection for EV and USN was 51.56 and 32.59 ng/band, while the limit of quantification was 156.23 and 98.76 ng/band, respectively. Intra- and interday precisions (n = 6) for EV and USN were 0.70-1.89 and 0.50-0.76 (%RSD), and 1.56-1.60 and 1.54-1.99 (%RSD), respectively. The mean percent recoveries were 99.66 and 99.87%, respectively, for EV and USN. However, USN was estimated in all four Usnea species but EV only in two species with varied quantity. Comparative antioxidant activity revealed that US is a better free radical scavenger in comparison with other three Usnea species. Furthermore, these results indicated that USN and EV are not solely responsible for antioxidant potential, but it may be due to synergistic effect.

Tinospora cordifolia (Thunb.) Miers, Menispermaceae, is a dioecious creeper, commonly known as "Giloe" or "Guduchi" with significant medicinal importance in the traditional systems of medicine. It is designated as Rasayana drug in Ayurveda and recommended for a number of diseases and also as adaptogen and immunomodulator. The safety and efficacy of herbal medicines are closely correlated with the quality of the source materials. The aim of this study is to see the effect of seasons on phytoconstituents and how these vary in male and female stem samples of T. cordifolia. The study revealed that total phenolics and total sugar concentration obtained highest values in summer season while starch and tannin content were found maximum in winter season in both the genders. However, biomarkers, tinosporaside and berberine, reached to their highest concentration in monsoon season. Further, antioxidant potential revealed the highest inhibition percentage in winter season as well as in late summer season. The results of this study suggest that the female plant is best for its therapeutic phytoconstituents and the best harvesting seasons may be either winter or late summer for antioxidant potential and immunomodulator activities and monsoon for antidiabetic activity of T. cordifolia.

ETHNOPHARMACOLOGICAL RELEVANCE: International organizations recognize the importance of sacred groves and place them into the context of sustainable development and also emphasize to conserve biodiversity through protection of sacred groves and sties. The significance of medicinal plants from Himalayan region is well known to the world. Therefore, present study was conducted in identified sacred groves of Kumaon Himalaya to investigate and document the utilization of medicinal plants by various local communities and tribal people. MATERIALS AND METHODS: The study was conducted during 2008-2011 in four seasons of the year. Information was collected from 70 locals from different sacred groves by using free listing interviews with randomly selected informants and semi-structured questionnaires; plant specimens were collected, identified and deposited at the CSIR-NBRI herbarium (LWG), Lucknow, India. RESULTS: Seven sacred groves viz., Dhwaj, Haat Kali, Hokra, Malay Nath, Nakuleshwar, Narayan Swami Ashram and Patal Bhuvneshwar were identified from the Pithoragarh district of Kumaon Himalaya. 89 medicinal plants belonging to 52 families and 77 genera of which, 2 are lichens, 4 are pteridophytes, 3 are gymnosperms and remaining 80 plant species are angiosperms. 6 plant species are reported with new therapeutic uses for the first time in this paper. Highest informant׳s consensus factor value was found in liver disorder (0.55) and least by body pains (0.23). 55 ethnomedicinal plants are showing 100% fidelity level against various diseases. CONCLUSION: Sacred groves in Kumaon region of Indian Himalaya are rich sources and best repository of ethno-medicinally important plants with many rare, endangered and threatened species. It is an excellent example of unique traditional way of in situ conservation of different plant species.

Catharanthus roseus (L.) G. Don, a medicinal plant, has a very important place in the traditional as well as modern pharmaceutical industry. Two common varieties of this plant rosea and alba are named so because of pink and white coloured flowers, respectively. This plant comprises of about 130 terpenoid indole alkaloids and two of them, vincristine and vinblastine, are common anticancer drugs. The effect of chromium (Cr) on enzymatic and non-enzymatic antioxidant components and on secondary metabolites vincristine and vinblastine was studied under pot culture conditions of both varieties of C. roseus. Antioxidant responses of these varieties were analyzed under 0, 10, 50, and 100 µM chromium (Cr) level in order to investigate the plant's protective mechanisms against Cr induced oxidative stress. The results indicated that Cr affects all the studied parameters and decreases growth performance. However, vincristine and vinblastine contents were increased under Cr stress. Results are quite encouraging, as this plant shows good antioxidant potential and increased the level of active constituents under Cr stress.

Bacopa monnieri (L.) is an important medicinal plant mainly known as a memory enhancing herb. It is important to see the effect of metal pollution on its active constituents. In this context, efforts have been made to observe the effect of Cd on the triterpenoid saponins bacoside A and bacopaside I in this plant. The influence of the metal on growth parameters like protein, chlorophyll content, and biomass has also been observed. It is interesting to note that the bacoside A and bacopaside I gradually increased by the Cd treatment up to 10 µM and then decreased at higher concentrations, that is, 50 and 100 µM, but the concentration of these components was more in all the treated plants as compared to control. On the contrary, protein, chlorophyll content, and biomass decreased with the increase in metal concentration and exposure duration due to metal toxicity.

Genus Adiantum L., Pteridaceae, is an important fern used in traditional systems of medicine. Different species of Adiantum are known as avenca in Brazil; hansraj/hanmspadi in India; maiden hair fern in English. The present study aims to develop the quality control parameters of four similar looking Adiantum species viz. A. capillus-veneris L., A. lunulatum Burm. f., A. peruvianum Klotzsch, and A. venustum D. Don. Standard methods for macro-microscopic evaluation, physico-chemical parameters and HPTLC were used for authentication and identification. The salient distinctive characters under the microscope are the presence of slightly wavy elongated epidermal cells in A. capillus-veneris; epidermal cells strongly wavy in A. lunulatum; star shaped epidermal cells and mixed spores of regular and irregular shaped in A. peruvianum; stomata on both the surfaces of pinnule, absence of spinulus spores only in A. venustum. In addition, rachis anatomy showed different cellular and stellar characteristics as identifying characters of aforesaid four Adiantum species. Physico-chemical parameters and HPTLC finger print profiles along with stigmasterol and lupeol play significant role for the quality evaluation of raw drugs. The above finding will serve the purpose of quality control and assurance for the future studies.

BACKGROUND: The stem barks are the rich sources of tannins and other phenolic compounds. Tannins inhibited the growth of various fungi, yeast, bacteria and virus. Hence, ten stem barks of ethnomedicinally important plants were screened for antibacterial and antifungal activities against human pathogenic strains. METHODS: Air-dried and powdered stem bark of each plant was extracted with 50% aqueous ethanol, lyophilized and the dried crude extracts were used for the screening against 11 bacteria and 8 fungi. Antibacterial and antifungal activities were performed according to microdilution methods by NCCLS. RESULTS: The plants Prosopis chilensis, Pithecellobium dulce, Mangifera indica showed significant antibacterial and antifungal activities against Streptococcus pneumonia, Enterobacter aerogenes, Klebsiella pneumonia and Candida albicans with MIC of 0.08mg/ml. Pithecellobium dulce bark also showed significant antibacterial activity against Bacillus cereus. CONCLUSION: The bark of Pithecellobium dulce has more or less similar activity against the known antibiotic and may be considered as potent antimicrobial agent for various infectious diseases.

Chromium and its compounds may cause disturbance in the nutrient level of the plants. Iron, manganese, copper, and zinc are essential nutrient elements and required for balanced growth and development of plants, but chromium uptake sometimes disturbed their concentration in plants. Therefore, in the present paper, an effort has been made to observe the effect of different levels of Cr on nutrient uptake of Phyllanthus amarus and Solanum nigrum, the medicinally important plants of indigenous systems of medicine having hepatoprotective and diuretic properties. The study revealed that Cr causes significant changes in nutrient uptake as compared to control plants. Besides, Cr-treated plants showed growth depression and decrease in fresh and dry weight too. With the increase in Cr supply, accumulation of Cr in roots was increased significantly. Concentration of manganese and zinc was also increased. However, copper concentration in both the plants seemed less affected by Cr.

Different species of Phyllanthus are considered to be very effective hepatoprotective agents in the Indian indigenous systems of medicine and are considered bitter, astringent, stomachic, diuretic, febrifuge, deobstruant and antiseptic. Still ayurvedic practitioners prescribed fresh juice of 'Bhuiamlki' for jaundice. Various species of Phyllanthus are being sold in India under the trade name 'Bhuiamlki'. During market surveillance of herbal drug, it was observed that almost all the commercial samples, either comprise of Phyllanthus amarus Schum & Thonn. or Phyllanthus maderaspatensis Linn. or mixture of Phyllanthus amarus, Phyllanthus fraternus Webster. and Phyllanthus maderaspatensis. Therefore, in this context the detailed pharmacognostical evaluation of all the three species has been carried out with the aim to establish the identification markers of this important hepatoprotective agent (effective in hepatitis B too). The study conclude that all the three species can be differentiated on the basis of macro and microscopic characters, physico-chemical values, HPTLC fingerprint profile, and detection of phyllanthin and hypophyllanthin as marker components. Besides, an interesting conclusion can also be drawn that phyllanthin and hypophyllanthin said to protect hepatocytes against carbon tetrachloride and galactosamine induced toxicity, may not be exclusively responsible for hepatoprotective activity as these are present only in Phyllanthus amarus while Phyllanthus fraternus and Phyllanthus maderaspatensis also possess significant hepatoprotective activity.

The pollution is increasing in the environment by different kinds of human activities, which results in the accumulation of heavy metals including cadmium in the soil and water and it causes different types of problems to living beings. As the plants are utilized by human being as food and medicine, therefore, it is mandatory to see the effect of metals on plants. In this context, efforts have been made to observe the effect of different concentration of Cadmium (Cd) on Phyllanthus amarus Schum. and Thonn., because Cd is the widespread metal and the plants response to low and high level of exposure is a complex phenomenon. P. amarus is mostly grown as weed in agricultural and waste lands. It is a reputed plant used in Indian indigenous systems of medicine with hepatoprotective, diuretic, stomachic properties and is recently being used for the treatment of hepatitis B. The study revealed that Cd causes significant decrease in fresh and dry weight, length of root and shoot, protein, chlorophyll, carotenoids and sugar and increase in starch content. It is interesting to note that the therapeutically active compounds-phyllanthin and hypophyllanthin, enhanced at certain levels of Cd due to abiotic stress. Besides, the ultramorpholical changes were also observed in stomatal opening and wax deposition on both the surfaces of leaves.
